Traditional ex vivo screening methods have failed to produce effective treatments, as age-related diseases have complex causes that include interactions with the aged environment. To address this problem, Gordian’s platform is the first and only to enable pooled in vivo tests at scale in our proprietary “patient avatars”. We are able to deliver and test hundreds of therapeutics in individual animals, and scale efficacy testing to cover every therapeutic target at the beginning of the drug discovery process (more info on our website). Specifically, you will architect experimental designs, set up new assays and evaluate the safety, efficacy, and mechanism of action of potential therapeutic targets. \n\n\nYour focus at Gordian is for the validation or elimination of screen hits to happen as early as possible. Gordian is unique in that our in vivo discovery delivers results at a far greater scale than even the largest pharma company. This means that we aren’t dependent on the success of a single target, but can survey the landscape of disease biology for the most promising target (or combination of targets) before moving to therapeutic development. You’ll work closely with our heads of disease biology, managing experimental schedules, and developing assays for in vivo and ex vivo workflows and setting up ex vivo systems to drive forward validation plans for our HFpEF and IPF programs. \n\n\nIn your first month you’ll become familiar with our technology and workflows, developing a strong understanding of the role of the preclinical team at Gordian. You’ll become proficient in our established ex vivo cultures and use your expertise to generate a plan for onboarding one new ex vivo system. Additionally, you’ll work with therapeutic area heads and consultants to generate plan for onboarding human precision cut lung and heart slices\n\n\nBy 2 months, you’ll have set up one complex ex vivo system, owning the entire project, both establishing robust culture methods and optimizing the delivery. You will also start to generate pilot data for a second ex vivo system.\n\n\nAt 3 months, you’ll generate SOPs for one ex vivo system and onboard and train teammates on the method. You’ll have established robust culture methods / delivery for the second ex vivo system. Additionally you’ll have initiated the validation of 10 targets in one ex vivo system as well and by month 4 you’ll have one the same in a second ex vivo system.\n\n\nBy 6 months, you’ll have completed the first round of target validation for two ex vivo systems. \n\n\nAnd You:\n\n\nYou want a chance to do your best work, immerse yourself and excel, alongside people who will inspire you and whom you’ll be excited to spend your days with. \n\n\nYou want to play a key role in an early-stage startup: A fast-paced environment full of both uncertainty and new challenges, demanding relentless resourcefulness.\n\n\nYou are excited to use cutting-edge technology to find the best therapeutic targets, and turn those into drugs. \n\n\nYou have experience in Mammalian cell culture with specific expertise in complex protocols (e.g. iPSC culture, slice cultures, organoids). \n\n\nIdeally you have done long term culture of heart tissues, human iPSCs and ESC culture and  differentiation. \n\n\nYou have experience in human ex vivo culture systems, specifically, with human cardiomyocytes and with human precision cut lung slices.\n\n\nYou’re experienced in RT-QPCR for gene expression in cell and tissue samples as well as Cryosectioning tissues. \n\n\nYou’re experienced with ELISAs, western blot, histology and image analysis (eg: ImageJ) and can effectively interpret the data results. \n\n\nYou have deep experience in assay development (experience with hydroxyproline assays is a plus).\n\n\nYou have experience managing and collaborating on time-sensitive projects, and are comfortable guiding and collaborating across multiple teams to execute experiments at a scale beyond any single person. \n\n\nExperience working in industry and working with Lentiviruses/AAVs are both pluses.\n\n\nThe Details:\n\n\nGordian aims to provide everything you need to thrive.
